From Slowing Aging to Reversing It: ER-100 Begins Human Trials Targeting Glaucoma and Optic Nerve Stroke
When doctors hear 'optic nerve damage' or 'late-stage glaucoma,' they often can only say, 'Vision can't be restored.' This is because retinal ganglion cells connect the eye to the brain, and once they die, they cannot regenerate. However, a groundbreaking development is emerging in the medical field. Life Biosciences in the US is advancing gene therapy ER-100, which has received FDA approval to begin its first human trials, aiming to turn back the eye's 'biological clock.' Traditional glaucoma treatment only lowers eye pressure, like hitting the brakes just before a car crashes into a wall. The damaged front of the car won't be restored, and nerve cells will continue to die. ER-100 doesn't aim to slow deterioration but to rejuvenate cells. It uses 'epigenetic reprogramming' to reset damaged retinal cells, akin to pressing a 'factory reset' button for aging and damaged cells. The team controls three transcription factors: OSK (OCT-4, SOX-2, KLF-4). These don't alter DNA but erase cellular 'aging markers,' turning back the biological clock. This makes damaged nerve cells mistakenly believe they are still young, thereby restoring their function. The therapy targets glaucoma and NAION, commonly known as eye stroke, for which there is currently no cure. In animal experiments, ER-100 has already helped non-human primates improve visual function. Phase I human trials will first confirm safety and immune response. If proven feasible, it will completely revolutionize how the medical community tackles degenerative diseases—moving beyond merely slowing aging to truly reversing it biologically. Not sure how long we'll have to wait for the experimental results, but if it's successful, it seems like it would be very useful for treatments, right? 🤔
